Mitochondrial respiratory chain complex I (NADH oxidase), mitochondrial respiratory chain complex II (succinate dehydrogenase) is a major element of electron entry into the mitochondrial electron transport chain (ETC). Complex I catalyzes the oxidation of NADH, and complex II catalyzes the oxidation of succinic acid to fumaric acid. Subsequently, coenzyme Q (Q) forms a coenzyme (QH2), which ultimately leads to a decrease in the terminal electron acceptor O2. Mitochondrial respiratory chain complex III (cytochrome c oxidoreductase) is an essential protein for mitochondrial oxidative phosphorylation. Mitochondrial respiratory chain complex III is the gatekeeper of the mitochondrial respiratory chain and the primary source of third reactive oxygen species. Mitochondrial respiratory chain complex IV (cytochrome c oxidase) is the terminal electron acceptor of the mitochondrial electron transport chain. Complex IV converts O2 to water by oxidation of cytochrome c, a process involved in ATP synthesis of mitochondrial cell membranes. The mitochondrial respiratory chain complex V, together with the above four complexes, completes oxidative phosphorylation to form ATP, called ATP synthase, also known as F1F0-ATPase.

Inhibition of mitochondrial respiratory chain complex activity leads to severe mitochondrial dysfunction, and long-term inhibition of mitochondrial respiratory chain complex activity may lead to neurological diseases such as Parkinson's disease, Down's syndrome, and Ricci syndrome. Principle display:
MitoCheck Complex I Activity Assay Kit (700930): reflects the activity of complex I by measuring the oxidation rate of NADH catalyzed by complex I. The oxidation rate of NADH is determined by the decrease in absorbance at 340 nm, and the decrease in absorbance is proportional to the activity of complex I.
MitoCheck Complex II Activity Assay Kit (700940): In complex II-catalyzed succinic acid oxidation, DCPIP was used as a staining agent, and the absorbance at 600 nm decreased with decreasing DCPIP. The activity of Complex II was determined by a decrease in absorbance at 600 nm.
