Brief introduction to fluorescence microscope operation

1. Boot:

1.1. Turn on the room main power switch

1.2. Turn on the computer power switch

1.3. Turn on the digital camera power switch

1.4. Turn on the microscope power switch (turn on the mercury lamp power switch)

2. Debug the microscope light path:

2.1. Place the slide on the stage. Adjust the focus.

2.2. Determine the aperture value of the condenser according to the objective index multiplied by 0.8 and adjust it to the position.

2.3. Reduce the field stop and then adjust the height of the concentrator until a clear image of the field stop is observed from the eyepiece.

2.4. Zoom in on the field diaphragm to extend the black frame in the eyepiece beyond the field of view.

3. Debug the digital camera to take photos:

3.1. Good field of view and focal length in the microscope viewfinder.

3.2. Open the photoshop program.

3.3. Click on "File - Auto - Kodakkms290acquine...." to bring up the camera control window.

3.4. ZOOM is set at 77. The picture format is 640*480 and do not change.

3.5. Click preview on the camera control window to observe the brightness of the preview image. Choose the appropriate exposure time to make the preview image look good.

3.6. Click the button to shoot, wait a few seconds and the picture will be sent back to the computer and displayed in the photoshop window.

3.7. Continue to take the next photo.

3.8. Save photos after taking ten photos.

3.9. Turn off the camera control button first. And save the shooting conditions to the default values.

3.10. Save the photo you just took to the specified folder in photoshop, and close the photo after saving.

3.11. Save and close all photos and reopen the camera control window to continue shooting.

3.12. Save the captured photos after the shooting is completed. Upload photos to the campus network server via the network when needed. You cannot use U disk or floppy disk to transfer files. Or burn the photo file to a disc.

4. Shutdown:

4.1. Turn off the microscope power.

4.2. Turn off the mercury lamp power supply.

4.3. Turn off the power of the digital camera.

4.4. When you turn off the computer, click "Restart the computer". When the computer enters the shutdown state and restarts, press the computer power switch to force shutdown. Note that you cannot directly click "Close Computer".

4.5. Turn off the main power switch of the room.
