Biological safety cabinet use and maintenance methods (1) Preparation work 1. Remove contamination from the inside surface of the biosafety cabinet before starting work.
2. Place all necessary items in the safety cabinet to minimize the number of times your arms enter and exit the front opening. Pay attention to the following points when placing items:
The intake grille on the front and rear of the Class 1 II biosafety countertop cannot be blocked by items.
2 All items should be placed as close as possible to the edge of the workbench at the rear of the workbench and will not block the rear grille during operation.
3 Equipment that produces aerosols (eg, mixers, centrifuges, etc.) should be placed close to the rear of the cabinet. Larger items such as biohazard waste bags and dishes containing discarded straws should be placed on one side of the safety cabinet.
4 Clean items should be placed at least 15 cm away from the aerosol vessel to reduce the possibility of cross-contamination.
5 Experimental samples susceptible to UV damage can be placed before the experiment after UV sterilization.
6 Do not discharge too many items in the work area.
3. Close the front window of the operation, open the UV lamp of the biosafety cabinet, and sterilize the work surface of the biosafety cabinet, the side wall surface and the inner side of the rear wall.
4. Open the front window slightly and let the biosafety cabinet work area vent for a few minutes before starting work.
(2) Operational precautions 1. Wear personal protective clothing and gloves to prevent contamination when using the biosafety cabinet. Ordinary lab coats can be worn when performing Level 1 and Level 2 biosafety operations. Gloves should be placed over the cuffs of the lab coat. Some operations may also require the use of masks and goggles.
2. In the biosafety cabinet, avoid using an open flame sterilizer such as an alcohol lamp. The use of open flames can have an effect on the airflow and can be dangerous when dealing with volatile and flammable materials. When sterilizing the inoculating loop, an infrared electric heater (see Figure 1) can be used, or a disposable plastic inoculating loop can be used.
3. Extend your hands and arms into the biosafety cabinet for about 1 minute, so that the safety cabinet can be adjusted and the air inside can be swept over the surface of the hands and arms before the items can be processed.
4. When working, try to reduce the movement of the arm, and move slowly to prevent interference with the airflow inside the cabinet. Care should be taken to maintain the integrity of the airflow in the front opening as you move your arms in and out of the safety cabinet. The arms should slowly enter and exit the front opening perpendicular to the opening in the work area.
5. The experimental operation on the work surface should be carried out in the direction from the cleaning zone to the contaminated zone. All operations are carried out in the middle and rear of the work surface.
6. When working, the autoclavable biohazard waste bag and the suction tube tray can not be taken out of the safety cabinet in the safety cabinet. Otherwise, the arms must frequently enter and exit the safety cabinet when using these items, which will interfere with safety. The integrity of the air barrier of the cabinet, which affects the protection of people and items.
7. Once the spill occurred during the experiment, place it on the surface with a sterile paper towel to remove it.
8. Try to keep the safety cabinet in continuous working condition and surface disinfection of all items to provide maximum personnel protection. When picking up items that may cause contamination from inside the cabinet, be sure to disinfect the surface.
(3) Processing after use 1. Fasten the used biological waste bag.
2. Clean and disinfect the inner wall, rear wall, work surface and inner side of the safety cabinet.
3. Since the remaining medium may cause microbial growth and reproduction, at the end of the experiment, all items including equipment should be cleaned of surface contamination and removed from the safety cabinet.
4. Before removing the arm from the safety cabinet, be sure to disinfect the surface and then slowly remove it perpendicular to the opening in the work area.
6. Remove the contamination of the inner surface of the biosafety cabinet after each use. The work surface and the inner wall should be wiped with a disinfectant, and the disinfectant used should be able to kill any microorganisms in the safety cabinet. At the end of each day's experiment, the surface of the biosafety cabinet, the surrounding area, and the inside and outside of the glass should be wiped to remove surface contamination. When effective against the target organism, it can be disinfected with a bleach solution or 70% alcohol. After using a corrosive disinfectant such as bleach, it must be wiped again with sterile water.
7. Close the front operating window and turn on the UV light.
(4) Disposal of spillage in the biosafety cabinet <br> Do not put the head into the safety cabinet when handling spillage, and do not face the front directly to the front operation port, but should be behind the front panel. The corrosive nature of the biosafety cabinet needs to be considered when selecting a sanitizer.
If the amount of spill is less than 1 ml, wipe it directly with a paper towel (or other material) soaked in disinfectant.
If the spill is large or the container is damaged, it is recommended to operate as follows: keep the biosafety cabinet open; cover the spill with the absorbent material impregnated with the disinfectant, and use it for a certain period of time to play the role of disinfection and sterilization. Soak the working surface with the disinfectant and the drainage ditch and the liquid receiving tank; after disinfecting and sterilizing the worn gloves in the safety cabinet, take off the gloves; if the protective clothing has been contaminated, take off the contaminated protective clothing, Wash the exposed area with a suitable sanitizer; wear appropriate personal protective equipment such as double gloves, protective clothing, goggles, and respiratory protection; carefully discard the spill (or other absorbent material) Collect the spilled material into a special collection bag or container and repeatedly use the new paper towel (or other absorbent material) to absorb the remaining material; the broken glass or other sharps should be treated with tweezers or pliers; Wipe or spray the inner wall of the safety cabinet, the working surface and the inside of the front window; after a certain period of time, wipe the disinfectant with clean water; if you need to soak the liquid tank, clean it The supervisor should be reported before the sump; it may need to be sterilized by other means before cleaning. If the spill is recycled into the biosafety cabinet, the appropriate measures need to be taken after the assessment.
(5) Regular maintenance 1. Replace the pre-filter every three months.
2. Replace the UV lamp once a year.
3. When one of the following conditions occurs, the biosafety cabinet should be inspected on site: after the biosafety laboratory is completed, the biosafety cabinet has been installed before being put into use; after the biosafety cabinet is moved, after the biosafety cabinet is inspected; After the biosafety cabinet is replaced with a high-efficiency filter; the biological safety cabinet is routinely tested annually. On-site testing shall be performed by qualified personnel in accordance with the manufacturer's instructions for the performance and integrity of each biosafety cabinet to verify compliance with national standards. Safety cabinet protection should be evaluated for safety cabinet integrity, HEPA filter leakage, vertical airflow speed, working window airflow and average wind speed, negative pressure/ventilation times, airflow smoke patterns, and alarms and interlocks. The system is tested. You can also choose to conduct leakage, illuminance, UV intensity, noise level, and vibration testing.
