Improved version of the viral vector can help gene therapy

Medical Network November 13th A European research team has recently developed a new method for the production of viral vectors, making viral vectors safer and more effective in transmitting related genes, which will help improve the clinical application of gene therapy.
As the name suggests, "viral vector" is the use of virus as a carrier, using the characteristics of the virus to transfer genes to cells, applied to gene therapy. The key to using this "tool" is safety and efficiency: you need to ensure that the virus does not harm the human body and can effectively transmit useful genes.
SV40 is a virus with high gene transfer efficiency and possesses the characteristics as a viral vector. However, the problem that has been difficult to solve in the past is that it is difficult to completely avoid the generation of wild virus when producing this viral vector, which limits its clinical application.
Researchers from the Netherlands and Spain reported in the latest issue of the journal Molecular Therapy that they have improved the production of SV40 viral vectors and improved safety on the basis of ensuring the effectiveness of their gene delivery.
Dr. Peter Hahn, one of the authors of the paper, explains that they use biotechnology to set up a double insurance for the virus vector. One is to ensure that there is no sequence between the DNA of the carrier and the receptor (deoxyribonucleic acid). Overlap, the second is to delete the STag gene directly related to the toxicity of this virus. "As a result, the probability of wild virus emergence during production is almost zero. Even if it occurs, the toxicity of the STag gene is greatly weakened. And it is very difficult to survive."
Researchers believe that this new approach will extend the application of viral vectors to help develop new gene therapies or vaccines.
