How to improve the accurate reproducibility of ELISA

The accuracy of the immunoassay is expressed as the repeatability between the single experimental wells (in-batch) and the repeatability between multiple experiments (inter-batch). A high CV value indicates low accuracy and is usually caused by two reasons: pipetting techniques and washing techniques.


Pipetting technology
1. When pipetting, the tip should be aligned with the center of the hole to avoid contact with the wall and bottom of the hole.
2. After pipetting, gently shake the mixing reagent.
3. If your sample is sticky, pre-clean the tip to eliminate the effects of surface tension. Wait for a while after aspiration to balance the volume before taking it out.
4. Insufficient pipetting or uneven pipetting volume indicates that the pipette needs to be calibrated. Check the pipette and recalibrate if necessary.
5. When loading the sample, ensure that the tip is replaced between different reagent standards and samples to avoid cross-contamination.
6. Make sure to use the appropriate tip. Unsuitable tips do not accurately measure the volume of aspiration and loading.


Washing technology
1. If using an automatic washer or multi-channel pipette, make sure the import and export are working properly.
2. After the last wash, turn the plate to pour out the excess wash solution and pat dry the liquid on the blotting paper. Washing the plate too fast or too slow will reduce the accuracy. Do not let the liquid in the hole dry naturally, and immediately proceed to the next step.
3. According to the instructions, add enough washing liquid to the hole and ensure the washing is complete.
4. Since the washing solution is not versatile, do not use the washing solution from other kits when the washing solution in the kit is used up. Contact technical support for assistance if needed.
5. Do not reduce the soaking time during washing or skip the soaking step.
6. Wash according to the number of washings recommended in the manual. Changing the number of washings at will affects the accuracy of the experiment.


Liquid storage bottle
To avoid contamination, different reagents use different reservoirs. Some reagents or samples are easily contaminated (such as saliva, oxidizer, etc.), please refer to the instructions. Take precautions to protect the solution when necessary.


Sealing film
The used sealer film may contain the reagents remaining in the previous step. If it is reused, the plate will be contaminated, resulting in low accuracy. It is recommended to use a new closure film for each incubation.
